Candidate Genes

Authors used novel data mining tool ExQuest to identify novel candidate genes for existing diabesity QTLs. Next, candidate gene expression in the liver, adipose, and pancreas of diabesity-prone Tally Ho mice and diabesity-resistant C57BL/6J mice was assessed by quantitative PCR analysis. Several potential candidate genes, some with no previous association to diabesity QTLs, were identified. Since QTL intervals may be large and could contain hundreds or thousands of potential candidate genes, this method allows researchers to focus on those with strong potential as well as identify novel candidate genes.

Potential candidate genes for QTLs Bw1 (18 cM) and Bw3 (32 cM) on mouse Chromosome X as identified by ExQuest are Rgn and Rab9, respectively.

Mapping and Phenotype information for this QTL, its variants and associated markers

By quantitative trait mapping using MAPMAKER/QTL the authors reported that regions on mouse Chromosome X were significantly associated with adult body weight in mice. Three loci, Bw1, Bw2 and Bw3 were mapped using two crosses.

In the first cross ((C3H/He x M. spretus))F1 females were mated to C57BL/6J males. In the first cross Bw1 mapped to a region defined proximally by DXMit57 and distally by DXMit48 and Bw2 by DXMit109 and DXMit16.

In the second cross ((A/J x M. spretus))F1 females were mated to C57BL/6J males. In this second cross Bw1 was mapped to a region defined proximally by DXNds1, Bw2 by DXMit60 and DXMit16 and Bw3 by DXMit3 and DXMit12.

01.07.2015 Curator Note: Because Bw1 and Bw2 were originally mapped here in the first cross, which differs from the second cross, we consider the second cross a separate mapping experiment and have named the QTL identified as Bw33 (defined proximally by DXNds1) and Bw34 (defined by DXMit60 and DXMit16), respectively.
