Name: v-raf-leukemia viral oncogene 1; targeted mutation 1, Andreas Zimmer
MGI ID: MGI:2429683
Synonyms: Craf-1neo2
Gene: Raf1  Location: Chr6:115618067-115676635 bp, - strand  Genetic Position: Chr6, 53.62 cM, cytoband C3
Germline Transmission:  Earliest citation of germline transmission: J:50696
Parent Cell Line:  MPI-II (ES Cell)
Strain of Origin:  129S2/SvPas
Allele Type:    Targeted (Hypomorph)
Mutation:    Insertion
Mutation detailsA hypomorphic allele was generated by disrupting the first coding exon (exon 2) with a neo cassette. While Western blot analysis showed an absence of normal protein (74 kDA), low levels of a 62 kDa protein were identified. Sequence analysis of mRNA showed excision of the targeted exon and identified potential start codons in exon 3. Biochemical assays showed no kinase activity in nonstimulated mutant fibroblasts and a 90% reduction in activity in stimulated fibroblasts relative to wild-type samples. (J:50696)
