Mutation detailsA 568 bp fragment of human PTHLH cDNA under the control of the mouse Col2a1 promoter was used as the transgene. The Col2a1 promoter sequences were used to target expressioin of the transgene to proliferating and prehypertrophic chondrocytes. An artificial exon was engineered into the 5' end of the Col2a1 construct to prevent alternative splicing and the translation initiation codon was inactivated to ensure translation of the human PTHLH gene. 2.2 kb of human growth hormone untranslated sequences were added downstream of PTHLH to provide termination and polyadenylation signals as well as increase expression efficiency. Transgenic animals express human PTHLH at approximately 4x the amount of endogenous mouse protein. (J:76983)
