Mapping and Phenotype information for this QTL, its variants and associated markers

QTLs associated with cocaine induced seizures were provisionally mapped in 24 BXD recombinant inbred (RI) strains (B = C57BL/6J, D = DBA/2J) and confirmed in a genome screen with (C57BL/6J x DBA/2J)F2 intercross animals. Animals were administered cocaine via tail vein injection and evaluated for running bouncing clonic (RBC) seizure and tonic hindlimb extension (THE) seizure. Parental strain C57BL/6J is more sensitive to cocaine induced THE seizures than parental strain DBA/2J, but there is no differencefor RBC seizures. Three significant loci and 2 suggestive loci were mapped and confirmed in the RI strains and (C57BL/6J x DBA/2J)F2 intercross. Cosz1 (in association with RBC) maps to distal mouse Chromosome 9 from 49 cM - 79 cM with a LOD = 3.6 near D9Mit51. DBA/2J-derived alleles at Cosz1 confer recessive resistance to seizures. Cosz1 maps near 2 other seizure-related QTLs, El1 and Bis3. A possible candidate for Cosz1 is Htr1b. Cosz2 (in association with THE) maps to mouse Chromosome 14 from 13 cM - 30 cM with a LOD = 5.1 between D14Mit54 and D14Mit39. DBA/2J-derived alleles confer additively inherited resistance to cocaine-induced seizures at Cosz2. A possible candidate gene for Cosz2 is Grid1. Cosz2 maps near El5, a QTL associated with partial epilepsy seizure. Cosz3 (in association with RBC and THE) maps to mouse Chromosome 15 from 51 cM - 69 cM with a LOD = 4.1 at D15Mit48. C57BL/6J-derived alleles confer additively inherited resistance to cocaine-induced seizures at Cosz3. Possible candidate genes for Cosz3 are Scn8a and Cacnb3 (Cchb3). Suggestive QTLs mapped to proximal mouse Chromosome 9 and distal mouse Chromosome 15. Loci on mouse Chromosome 9 appear to be female-specific.
