Mutation details: The 166 kb C57BL/6J mouse bacterial artificial chromosome (BAC) from the RPCI23 library containing the entire Pax8 locus (and other genes) was modified by targeting a yellow fluorescent protein (YFP) coding sequence into the ATG start site of Pax8. This inserted a YFP fusion protein coding sequence, SV40 polyA signal, and a FRT-flanked Kanamycin resistance gene cassette. No other genes on the BAC were modified or deleted. Flp-mediated recombination removed the Kanamycin cassette. The BAC clone backbone vector (pBACe3.6), which contains the chloramphenicol resistance (CmR) gene, was not removed prior to microinjection and thus is retained in the transgenic animals.
