Mutation detailsThe transgene contains a cDNA encoding human growth differentiation factor 15 (GDF15; also called NAG-1, nonsteroidal anti-inflammatory drug-activated gene) downstream of the "CAG promoter," which consists of the cytomegalovirus immediate early (CMV-IE) enhancer followed by the promoter, first exon and first intron of the chicken beta-actin gene, with the 3' splice junction sequence replaced by that of the rabbit hemoglobin beta gene. Immunoblot analysis with a rabbit antiserum recognizing human, but not mouse, GDF15 protein demonstrated high expression levels in skin, kidney, colon and brain, lower levels in lung, and no detectable expression in liver of transgenic mice.
