Mutation detailsA human cDNA encoding the short isoform (1072 amino acids) of RET was mutated by PCR to introduce the RET MEN2B mutation (Met918Thr). This mutated cDNA was inserted downstream of a human DBH promoter (5.8 kb). Between the transcriptional start site and the mutated cDNA, the construct contains the first intron of the rat preproinsulin gene (+13 +183 nt). A polyadenylation site from the murine protamine 1 gene was inserted at the 3' end. This transgenic line has a copy number of 70. (J:108163)
